This variant lies in the PALB2 gene (transcript NM_024675.4) at coding-DNA position 1395, where G is replaced by A; at the protein level this means replaces methionine at residue 465 with isoleucine — a missense variant. Submitter rationale: This variant has not been reported in the literature in individuals with PALB2-related disease. ClinVar contains an entry for this variant (Variation ID: 186522). Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0".
