This variant lies in the BRCA2 gene (transcript NM_000059.4) at coding-DNA position 3683, where A is replaced by G; at the protein level this means replaces asparagine at residue 1228 with serine — a missense variant. Submitter rationale: The p.N1228S variant (also known as c.3683A>G), located in coding exon 10 of the BRCA2 gene, results from an A to G substitution at nucleotide position 3683. The asparagine at codon 1228 is replaced by serine, an amino acid with highly similar properties. This variant was identified in 1 of 2351 breast and/or ovarian cancer patients undergoing BRCA1/2 genetic testing (Santonocito C et al. Cancers (Basel), 2020 May;12:). This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
