Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Color Diagnostics, LLC DBA Color Health to NM_000038.6(APC):c.6028A>G (p.Lys2010Glu), citing ACMG Guidelines, 2015: This missense variant replaces lysine with glutamic acid at codon 2010 of the APC protein. This is a missense variant not located in the first 15-amino acid repeat of the beta--catenin binding domain (codon 1021-1035) (PMID: 37800450). To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with APC-related disorders in the literature. This variant has been identified in 9/1613320 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
