Uncertain significance for Familial cancer of breast; Fanconi anemia complementation group J — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_032043.3(BRIP1):c.1362A>C (p.Glu454Asp), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the BRIP1 gene (transcript NM_032043.3) at coding-DNA position 1362, where A is replaced by C; at the protein level this means replaces glutamic acid at residue 454 with aspartic acid — a missense variant. Submitter rationale: This variant has not been reported in the literature in individuals with BRIP1-related conditions. ClinVar contains an entry for this variant (Variation ID: 185909). This sequence change replaces glutamic acid with aspartic acid at codon 454 of the BRIP1 protein (p.Glu454Asp). The glutamic acid residue is weakly conserved and there is a small physicochemical difference between glutamic acid and aspartic acid. This variant is not present in population databases (ExAC no frequency). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
