NM_002878.4(RAD51D):c.532A>G (p.Met178Val) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.M178V variant (also known as c.532A>G), located in coding exon 6 of the RAD51D gene, results from an A to G substitution at nucleotide position 532. The methionine at codon 178 is replaced by valine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
