Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_005732.4(RAD50):c.2260C>A (p.Leu754Met), citing Ambry Variant Classification Scheme 2023: The p.L754M variant (also known as c.2260C>A), located in coding exon 14 of the RAD50 gene, results from a C to A substitution at nucleotide position 2260. The leucine at codon 754 is replaced by methionine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
