Pathogenic for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000179.3(MSH6):c.397_410del (p.Phe133fs), citing Ambry Variant Classification Scheme 2023: The c.397_410del14 pathogenic mutation, located in coding exon 2 of the MSH6 gene, results from a deletion of 14 nucleotides at nucleotide positions 397 to 410, causing a translational frameshift with a predicted alternate stop codon (p.F133Pfs*7). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
