Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_024675.4(PALB2):c.40A>G (p.Lys14Glu), citing Ambry Variant Classification Scheme 2023: The p.K14E variant (also known as c.40A>G), located in coding exon 1 of the PALB2 gene, results from an A to G substitution at nucleotide position 40. The lysine at codon 14 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
