NM_007294.4(BRCA1):c.1042T>C (p.Cys348Arg) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.C348R variant (also known as c.1042T>C), located in coding exon 9 of the BRCA1 gene, results from a T to C substitution at nucleotide position 1042. The cysteine at codon 348 is replaced by arginine, an amino acid with highly dissimilar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
