Uncertain significance for Malignant tumor of breast — the classification assigned by Department of Pathology and Laboratory Medicine, Sinai Health System to NM_000465.4(BARD1):c.2091A>T (p.Ala697=): The BARD1 p.Ala697= variant was not identified in the literature nor was it identified in the Cosmic, MutDB, or Zhejiang University databases. The variant was identified in dbSNP (ID: rs786201573) as "With Likely benign allele", and in ClinVar (classified as likely benign by Ambry Genetics, Invitae). The variant was not identified in the following control databases: the 1000 Genomes Project, the NHLBI GO Exome Sequencing Project, the Exome Aggregation Consortium (August 8th 2016), or the Genome Aggregation Database (Feb 27, 2017). The p.Ala697= variant is not expected to have clinical significance because it does not result in a change of amino acid and is not located in a known consensus splice site. The variant occurs outside of the splicing consensus sequence but 4 of 4 in silico or computational prediction software programs (SpliceSiteFinder, MaxEntScan, NNSPLICE, GeneSplicer) predict a greater than 10% chance that a cryptic splice site located at this loci is lost due to this variation.
