Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Sema4, Sema4 to NM_001048174.2(MUTYH):c.1107G>T (p.Leu369=), citing Sema4 Curation Guidelines. This variant lies in the MUTYH gene (transcript NM_001048174.2) at coding-DNA position 1107, where G is replaced by T; at the protein level this means the protein sequence is unchanged (leucine at residue 369 retained) — a synonymous variant. Submitter rationale: The MUTYH c.1191G>T (p.L397=) variant has not been reported in the literature to our knowledge. It was observed in 14/128332 chromosomes of the Non-Finnish European subpopulation, including no homozygotes, in the large and broad cohorts of the Genome Aggregation Database (http://gnomad.broadinstitute.org, PMID: 32461654). The variant has been reported in ClinVar (Variation ID 184102). In silico tools suggest that the variant may have an impact on splicing, though these predictions have not been confirmed by functional studies. The evidence is insufficient to meet ACMG/AMP criteria for classifying the variant as benign or pathogenic. Thus, the clinical significance of this variant is currently uncertain.
