NM_007194.4(CHEK2):c.296T>C (p.Leu99Pro) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.L99P variant (also known as c.296T>C), located in coding exon 1 of the CHEK2 gene, results from a T to C substitution at nucleotide position 296. The leucine at codon 99 is replaced by proline, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be benign and deleterious by PolyPhen and SIFT in silico analyses, respectively. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
