NM_000455.5(STK11):c.950A>C (p.Glu317Ala) was classified as Uncertain significance by GeneDx, citing GeneDx Variant Classification (06012015): This variant is denoted STK11 c.950A>C at the cDNA level, p.Glu317Ala (E317A) at the protein level, and results in the change of a Glutamic Acid to an Alanine (GAA>GCA). This variant has not, to our knowledge, been published in the literature as pathogenic or benign. STK11 Glu317Ala was not observed in approximately 6,500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. Since Glutamic Acid and Alanine differ in polarity, charge, size or other properties, this is considered a non-conservative amino acid substitution. STK11 Glu317Ala occurs at a position that is conserved across species and is not located in a known functional domain (UniProt). In silico analyses are inconsistent regarding the effect this variant may have on protein structure and function. Based on currently available information, it is unclear whether STK11 Glu317Ala is pathogenic or benign. We consider it to be a variant of uncertain significance.
