Submitter rationale: Variant summary: MLH1 c.1243G>A (p.Asp415Asn) results in a conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be tolerated. The variant allele was found at a frequency of 1.6e-05 in 251316 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.1243G>A has been observed in individuals affected with Brease Cancer or Hereditary cancer predisposition syndromes without unequivocal evidence for causality (Tung_2015, Bhai_2021). These reports do not provide unequivocal conclusions about association of the variant with Lynch Syndrome.
