Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Sema4, Sema4 to NM_004655.4(AXIN2):c.1948C>T (p.Arg650Cys), citing Sema4 Curation Guidelines. This variant lies in the AXIN2 gene (transcript NM_004655.4) at coding-DNA position 1948, where C is replaced by T; at the protein level this means replaces arginine at residue 650 with cysteine — a missense variant. Submitter rationale: The AXIN2 c.1948C>T (p.R650C) variant has not been reported in literature to our knowledge. It was observed in 12/24668 chromosomes in the African/African American population, according to the Genome Aggregation Database (http://gnomad.broadinstitute.org, PMID: 32461654). This variant has been reported in ClinVar (Variation ID 182009). Functional studies have not been performed and in silico tool predictions of the variants effect on protein function are inconclusive. The evidence is insufficient to meet ACMG/AMP criteria for classifying the variant as benign or pathogenic. Thus, the clinical significance of this variant is currently uncertain.
