The region that overlaps with the duplication area is the LEMD3 gene.The LEMD3 gene acts as a particular TGF-beta, activin, and BMP signalling regulator by interfering with the R-SMAD protein complexes. Its antagoniser TGF-beta induced cell proliferation arrest. It can therefore be assumed that the copy number gain in LEMD3 interrupts the TGFβ signalling function in regulating palate development in both epithelium and mesenchyme, thus leading to the formation of CL/P (Iwata et al., 2011)
