This variant lies in the MYBPC3 gene (transcript NM_000256.3) at coding-DNA position 1201, where C is replaced by T; at the protein level this means converts the codon for glutamine at residue 401 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: p.Gln401Stop (CAG>TAG): c.1201 C>T in exon 13 of the MYBPC3 gene (NM_000256.3)The Gln401Stop mutation in the MYBPC3 gene has not been reported as a disease-causing mutation or as a benign polymorphism to our knowledge. Gln401Stop is predicted to cause loss of normal protein function either by protein truncation or nonsense-mediated mRNA decay. Other nonsense mutations in the MYBPC3 gene have been reported in association with HCM. In summary, Gln401Stop in the MYBPC3 gene is interpreted as a disease-causing mutation.
