The E843K variant has not been published as pathogenic or been reported as benign to our knowledge. The E843K variant is not observed in large population cohorts (Lek et al., 2016). The E843K variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. Furthermore, in-silico analyses, including protein predictors and evolutionary conservation, support a deleterious effect. However, although missense variants in nearby residues have been reported in the Human Gene Mutation Database, the clinical significance of these variants is currently unknown (Stenson et al., 2014).
