pathogenic — the classification assigned by Quest Diagnostics Nichols Institute San Juan Capistrano to NM_007294.3:c.5075_5193del, citing Quest Diagnostics criteria: A deletion of exons 17-18 (c.5075_5193, also known as exons 18-19 in legacy exon nomenclature) in the BRCA1 gene is predicted to disrupt the translation reading frame of the BRCA1 mRNA and result in the premature termination of BRCA1 protein synthesis. In the published literature, a similar deletion of exons 17-18 in the BRCA1 gene has been reported in individuals with breast and/or ovarian cancer (PMID: 12700174 (2003), 17561994 (2007), 17333342 (2007), 34254208 (2021), 34413315 (2021)). This variant has not been reported in large, multi-ethnic general populations (Genome Aggregation Database, http://gnomad.broadinstitute.org). Based on the available information, this variant is classified as pathogenic.
