NM_024675.3:c.2835_3113del was classified as likely pathogenic by Quest Diagnostics Nichols Institute San Juan Capistrano, citing Quest Diagnostics criteria: A deletion of exons 9-10 (c.2835_3113) in the PALB2 gene is predicted to maintain the translation reading frame of the PALB2 mRNA, however, it removes a portion of the gene important for its structure or function (PMID: 24141787 (2014)). In the published literature, similar deletions of exons 9-10 in the PALB2 gene have been reported in individuals/families affected with breast/ovarian cancer (PMID: 24136930 (2013), 32624572 (2020), 37686625 (2023)). This variant has not been reported in large, multi-ethnic general populations (Genome Aggregation Database, http://gnomad.broadinstitute.org). Based on the available information, this variant is classified as likely pathogenic.
