Pathogenic — the classification assigned by Quest Diagnostics Nichols Institute San Juan Capistrano to GRCh37/hg19 17p12(chr17:14073563-15479923)x3, citing ACMG/ClinGen CNV Guidelines, 2019: This copy number gain includes the PMP22 gene (OMIM 601097) and is consistent with Charcot-Marie-Tooth disease type 1A (OMIM 118220). Duplication of PMP22 accounts for 70-80% of all Charcot-Marie-Tooth neuropathy type 1 (CMT1), a demyelinating peripheral neuropathy characterized by distal muscle weakness, atrophy, sensory loss, and slow nerve conduction velocity. The penetrance of this duplication is thought to be near 100%; however, age of onset and severity of the condition are variable, and some carriers are not clinically recognized.
