Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_005159.5(ACTC1):c.10G>C (p.Asp4His), citing Ambry Variant Classification Scheme 2023: The p.D4H variant (also known as c.10G>C), located in coding exon 1 of the ACTC1 gene, results from a G to C substitution at nucleotide position 10. The aspartic acid at codon 4 is replaced by histidine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
