Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_000435.3(NOTCH3):c.770C>T (p.Thr257Ile), citing Ambry Variant Classification Scheme 2023: The c.770C>T (p.T257I) alteration is located in exon 5 (coding exon 5) of the NOTCH3 gene. This alteration results from a C to T substitution at nucleotide position 770, causing the threonine (T) at amino acid position 257 to be replaced by an isoleucine (I). Based on data from gnomAD, the T allele has an overall frequency of 0.003% (1/31404) total alleles studied. The highest observed frequency was 0.007% (1/15434) of European (non-Finnish) alleles. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
