NM_000388.4(CASR):c.2306C>G (p.Ser769Cys) was classified as Uncertain significance for Nephrolithiasis/nephrocalcinosis by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S769C variant (also known as c.2306C>G), located in coding exon 6 of the CASR gene, results from a C to G substitution at nucleotide position 2306. The serine at codon 769 is replaced by cysteine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
