Pathogenic — the classification assigned by Athena Diagnostics to NM_014231.5(VAMP1):c.340+2T>A, citing Athena Diagnostics Criteria. This variant lies in the VAMP1 gene (transcript NM_014231.5) at the canonical splice donor site of the intron immediately after coding-DNA position 340, where T is replaced by A; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: This variant is expected to severely impact normal RNA splicing, and consequently, protein structure and/or function. This variant occurs at the same position as another variant considered to be pathogenic (PMID: 22958904), strongly indicating this variant may also cause disease. This variant has been identified in at least one individual tested at Athena Diagnostics with clinical features associated with this gene. This variant has not been reported in large, multi-ethnic general populations (http://gnomad.broadinstitute.org).
