Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_000071.3(CBS):c.209C>G (p.Pro70Arg), citing Ambry Variant Classification Scheme 2023: The p.P70R variant (also known as c.209C>G), located in coding exon 1 of the CBS gene, results from a C to G substitution at nucleotide position 209. The proline at codon 70 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
