Submitter rationale: Variant summary: HPS4 c.1330G>A (p.Glu444Lys) results in a conservative amino acid change in the encoded protein sequence. Five of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 5.2e-05 in 250994 control chromosomes (gnomAD). This frequency is not significantly higher than expected for a pathogenic variant in HPS4 causing Hermansky-Pudlak Syndrome (5.2e-05 vs 0.00052), allowing no conclusion about variant significance.
