This variant lies in the WDR35 gene (transcript NM_020779.4) at the canonical splice acceptor site of the intron immediately before coding-DNA position 437, where G is replaced by A; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: Variant summary: WDR35 c.437-1G>A is located in a canonical splice-site and is predicted to affect mRNA splicing resulting in a significantly altered protein due to either exon skipping, shortening, or inclusion of intronic material. Several computational tools predict a significant impact on normal splicing: Four predict the variant abolishes a 3 prime acceptor site. Three predict the variant creates a 3 prime acceptor site. However, these predictions have yet to be confirmed by functional studies. The variant was absent in 251328 control chromosomes.
