Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_001354712.2(THRB):c.980C>T (p.Thr327Ile), citing LabCorp Variant Classification Summary - May 2015: Variant summary: THRB c.980C>T (p.Thr327Ile) results in a non-conservative amino acid change located in the Nuclear hormone receptor, ligand-binding domain (IPR000536) of the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 251464 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.980C>T has been reported in the literature in at-least one individual affected with Thyroid Hormone Resistance, Generalized, Autosomal Dominant and also in an unaffected father (example, Lee_2014, cited in Choi_2018). These data do not allow any conclusion about variant significance. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. No clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014. Based on the evidence outlined above, the variant was classified as uncertain significance.
