This variant lies in the ABCB4 gene (transcript NM_000443.4) at coding-DNA position 212, where T is replaced by A; at the protein level this means replaces leucine at residue 71 with histidine — a missense variant. Submitter rationale: Variant summary: ABCB4 c.212T>A (p.Leu71His) results in a non-conservative amino acid change located in the ABC transporter type 1, transmembrane domain (IPR011527) of the encoded protein sequence. Three of four in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 4e-06 in 251332 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.212T>A has been reported in the literature in a heterozygous individual affected with low-phospholipid-associated cholelithiasis syndrome (example: Poupon_2013) . This report does not provide unequivocal conclusions about association of the variant with Familial Intrahepatic Cholestasis. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
