Uncertain significance for Progressive external ophthalmoplegia with mitochondrial DNA deletions, autosomal recessive 1; Progressive sclerosing poliodystrophy; Mitochondrial DNA depletion syndrome 4b; Sensory ataxic neuropathy, dysarthria, and ophthalmoparesis — the classification assigned by Diagnostics Services (NGS), CSIR - Centre For Cellular And Molecular Biology to NM_002693.3(POLG):c.1403A>T (p.Asn468Ile), citing ACMG Guidelines, 2015: The c.1403A>T variant is not present in publicly available population databases like 1000 Genomes, EVS, ExAC, gnomAD, Indian Exome Database or our in-house exome database. This variant has neither been published in literature nor reported to clinical databases like ClinVar, Human Genome Mutation Database (HGMD) and OMIM. In-silico pathogenicity prediction programs like SIFT, PolyPhen-2, MutationTaster2, CADD etc. predicted this variant to be likely deleterious, however these predictions were not confirmed by any published functional studies.
