Uncertain significance for Dilated cardiomyopathy 1V — the classification assigned by Diagnostics Services (NGS), CSIR - Centre For Cellular And Molecular Biology to NM_000447.3(PSEN2):c.250G>A (p.Gly84Arg), citing ACMG Guidelines, 2015. This variant lies in the PSEN2 gene (transcript NM_000447.3) at coding-DNA position 250, where G is replaced by A; at the protein level this means replaces glycine at residue 84 with arginine — a missense variant. Submitter rationale: The c.250G>A variant is not present in publicly available population databases like 1000 Genomes, Exome Variant Server (EVS), Exome Aggregation Consortium (ExAC) and Indian Exome Database. The heterozygous state of the variant is present in Genome Aggregation Database (gnomAD), at a very low frequency. The variant is not present in our in-house exome database. The variant was not previously reported to ClinVar, Human Genome Mutation Database (HGMD) and/or OMIM databases, in any affected individuals. In-silico pathogenicity prediction programs like SIFT, Polyphen-2, MutationTaster2, CADD etc. predicted this variant to be likely deleterious, however these predictions were not confirmed by any published functional studies.
