This variant lies in the SRD5A2 gene (transcript NM_000348.4) at coding-DNA position 10, where C is replaced by T; at the protein level this means converts the codon for glutamine at residue 4 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: The c.10C>T variant is not present in publicly available population databases like 1000 Genomes, Exome Variant Server (EVS), Exome Aggregation Consortium (ExAC), Genome Aggregation Database (gnomAD) and Indian Exome Database. The variant is not present in our in-house exome database. The variant was not previously reported to ClinVar, Human Genome Mutation Database (HGMD) and/or OMIM databases, in any affected individuals. In-silico pathogenicity prediction programs like MutationTaster2, CADD, Varsome etc. predicted this variant to be likely deleterious. The variant creates a premature translational stop codon at the 4th amino acid position of the original transcript and expected to cause nonsense mediated decay of the mRNA.
