NM_002354.3(EPCAM):c.214G>A (p.Glu72Lys) was classified as Uncertain significance for Lynch syndrome 8; Breast carcinoma by Foundation for Research in Genetics and Endocrinology, FRIGE's Institute of Human Genetics, citing ACMG Guidelines, 2015: A heterozygous missense variation in exon 3 of the EPCAM gene that results in the amino acid substitution of Lysine for Glutamic Acid at codon 72 was detected. The p.Glu72Lys variant has not been reported in the 1000 genomes and gnomAD databases . The in silico predictions of the variant are probably damaging by PolyPhen-2 (HumDiv), damaging by SIFT, LRT and Mutation Taster2 tools. The reference codon is conserved across species. In summary, the variant meets our criteria to be classified as a variant of uncertain significance.
