NM_004990.4(MARS1):c.2278dup (p.Met760fs) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The c.2278dupA variant, located in coding exon 18 of the MARS gene, results from a duplication of A at nucleotide position 2278, causing a translational frameshift with a predicted alternate stop codon (p.M760Nfs*5). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. However, loss of function of MARS has not been clearly established as a mechanism of disease. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
