Uncertain significance — the classification assigned by Ambry Genetics to NM_139076.3(ABRAXAS1):c.13A>C (p.Ser5Arg), citing Ambry Variant Classification Scheme 2023: The p.S5R variant (also known as c.13A>C), located in coding exon 1 of the FAM175A gene, results from an A to C substitution at nucleotide position 13. The serine at codon 5 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
