Uncertain significance — the classification assigned by Ambry Genetics to NM_001008537.3(NEXMIF):c.2099T>A (p.Val700Glu), citing Ambry Variant Classification Scheme 2023: The p.V700E variant (also known as c.2099T>A), located in coding exon 2 of the KIAA2022 gene, results from a T to A substitution at nucleotide position 2099. The valine at codon 700 is replaced by glutamic acid, an amino acid with dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
