Uncertain significance — the classification assigned by Ambry Genetics to NM_002047.4(GARS1):c.308G>C (p.Arg103Thr), citing Ambry Variant Classification Scheme 2023: The p.R103T variant (also known as c.308G>C), located in coding exon 2 of the GARS gene, results from a G to C substitution at nucleotide position 308. The arginine at codon 103 is replaced by threonine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
