NM_003640.5(ELP1):c.67T>C (p.Cys23Arg) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.C23R variant (also known as c.67T>C), located in coding exon 1 of the IKBKAP gene, results from a T to C substitution at nucleotide position 67. The cysteine at codon 23 is replaced by arginine, an amino acid with highly dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
