NM_018076.5(ODAD2):c.251T>C (p.Val84Ala) was classified as Uncertain significance for Primary ciliary dyskinesia by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.V84A variant (also known as c.251T>C), located in coding exon 2 of the ARMC4 gene, results from a T to C substitution at nucleotide position 251. The valine at codon 84 is replaced by alanine, an amino acid with similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
