Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_012472.6(DNAAF11):c.106C>G (p.Leu36Val), citing Ambry Variant Classification Scheme 2023: The p.L36V variant (also known as c.106C>G), located in coding exon 2 of the LRRC6 gene, results from a C to G substitution at nucleotide position 106. The leucine at codon 36 is replaced by valine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
