NM_005591.4(MRE11):c.356_357insCAA (p.Ser119_Ile120insLys) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023. This variant lies in the MRE11 gene (transcript NM_005591.4) at coding-DNA position 356 through coding-DNA position 357, inserting CAA. Submitter rationale: The c.356_357insCAA variant (also known as p.S119_I120insK), located in coding exon 4 of the MRE11A gene, results from an in-frame CAA insertion at nucleotide positions 356 to 357. This results in the insertion of an extra lysine residue between codons 119 and 120. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis (Choi Y et al. PLoS ONE. 2012; 7(10):e46688). Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
