Uncertain significance — the classification assigned by Ambry Genetics to NM_004990.4(MARS1):c.715G>A (p.Ala239Thr), citing Ambry Variant Classification Scheme 2023: The p.A239T variant (also known as c.715G>A), located in coding exon 7 of the MARS gene, results from a G to A substitution at nucleotide position 715. The alanine at codon 239 is replaced by threonine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
