NM_005591.4(MRE11):c.187C>A (p.His63Asn) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.H63N variant (also known as c.187C>A), located in coding exon 3 of the MRE11A gene, results from a C to A substitution at nucleotide position 187. The histidine at codon 63 is replaced by asparagine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
