NM_004990.4(MARS1):c.2392-5dup was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The c.2392-5dupT intronic variant results from a duplication of two nucleotides upstream from coding exon 19 of the MARS gene. This nucleotide position is poorly conserved in available vertebrate species. In silico splice site analysis predicts that this alteration will not have any significant effect on splicing. Based on the supporting evidence, this variant is unlikely to be causative of axonal Charcot-Marie-Tooth disease, type 2U (CMT2U); however, its contribution to the development of interstitial lung and liver disease is uncertain.
