Uncertain significance — the classification assigned by Ambry Genetics to NM_003640.5(ELP1):c.221T>C (p.Val74Ala), citing Ambry Variant Classification Scheme 2023: The p.V74A variant (also known as c.221T>C), located in coding exon 2 of the IKBKAP gene, results from a T to C substitution at nucleotide position 221. The valine at codon 74 is replaced by alanine, an amino acid with similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
