Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_001211.6(BUB1B):c.3065T>A (p.Val1022Asp), citing Ambry Variant Classification Scheme 2023: The p.V1022D variant (also known as c.3065T>A), located in coding exon 23 of the BUB1B gene, results from a T to A substitution at nucleotide position 3065. The valine at codon 1022 is replaced by aspartic acid, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
