Uncertain significance — the classification assigned by Ambry Genetics to NM_004336.5(BUB1):c.3060A>C (p.Arg1020Ser), citing Ambry Variant Classification Scheme 2023: The p.R1020S variant (also known as c.3060A>C), located in coding exon 24 of the BUB1 gene, results from an A to C substitution at nucleotide position 3060. The arginine at codon 1020 is replaced by serine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
