NM_000075.4(CDK4):c.304A>G (p.Thr102Ala) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.T102A variant (also known as c.304A>G), located in coding exon 2 of the CDK4 gene, results from an A to G substitution at nucleotide position 304. The threonine at codon 102 is replaced by alanine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
