NM_002519.3(NPAT):c.3038C>T (p.Ser1013Leu) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S1013L variant (also known as c.3038C>T), located in coding exon 16 of the NPAT gene, results from a C to T substitution at nucleotide position 3038. The serine at codon 1013 is replaced by leucine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
